LOT 2 SPECIFICATIONS FOR JOHNE’S DISEASE
ELISA TEST KITS
1. Test kit must be approved and licensed by the USDA Veterinary Biologics.
2. Conjugate must be provided in a ready to use form that is stable for long term.
3. Chromogen solution must be provided in a ready to use form that is stable for long term.
4. Positive and negative control serum must be pre-absorbed and not handled identical to the
test sera.
5. The stop solution must be added to the ELISA plates after a 15 minute incubation. |

LOT 1 SPECIFICATIONS FOR JOHNE’S DISEASE
ELISA TEST KITS
1. Test kit must be approved and licenses by USDA Veterinary Biologics.
2. Conjugate must be provided in a concentrated solution and not ready to use. Ready to use conjugate is not as stable for long term storage as concentrated conjugate.
3. Positive and negative control serum provided by the manufacturer must be handled identical to test sera. For Johne’s disease this means the serum must be added to the Mycobacterium phlei diluent prior to adding to the antigen coated ELISA plates.
4. If the negative control serum is not added to the M. phlei diluent, it must yield an invalid test result because the optical density value for the negative control is too high. This is a critical internal control to monitor the effectiveness of the absorption step to remove cross-reactive antibodies.
5. The repeatability between ELISA plates for the same diagnostic samples must have an average coefficient of variation of 15% or less.
6. The stop solution must be added to the ELISA plates when the positive control wells reaches a designated optical density value. This assures the assay is optimized and follows first order kinetics. |
